I want to go ahead with purchasing the full screen activation apple carplay from: https://bimmer-remote.com/products/firmware-upgrade-for-toyota-supra-carplay-fullscreen

This might be a silly question, throughout the firmware update process, will it lock my dme?

Yes, but it's an unlockable lock, it's not like the ununlockable lock on the July 2020 and later ECUs.

Unlocking the ECU will cost more than not getting the recall and buying the full screen carplay yourself, but then you don't have the brake fix, so it's your call.

To answer your question, no, upgrading the display firmware for full screen carplay won't lock the ECU or affect any tune you've flashed.
