how many gallons of e85 are you guys putting in your car at empty to reach e50?

The Supra has a 13.7 gallon tank. Disregarding that, if you mix 50/50 e85 with 91/10 then you get roughly e48. If you mix 5.5 e85 and 4.5 91/10 you get just over e50. You would want to test the gas though as it could be as low as e70 in some places which means a 50/50 mix is now e40.

If you plan on running E50 you should get a flexfuel kit.
After a while it will be impossible to know your E level in your gas tank.
Why? Because the level of E in E85 varies and you seldom run your tank completely empty which means after a while there is no way of knowing the exact E level you got. The various apps only work correct if the E level is constant, which it is not.
With a flexfuel kit the ECU will handle timing etc.

Like previously mentioned, it becomes tricky due to ethanol pumps not always having exactly 85% ethanol. I went with a flex fuel kit and tune to remove the necessity of being precise and to not worry about map switching (1 tune will auto adjust to ethanol content). But regardless, it can still be a bit of a guessing game until you get readout from the sensor after a full up or perform a test on that batch of fuel.

In California pump e85 is exempt from the most recent road tax levied because it is considered a green fuel. As such the ethanol content is tightly regulated and is always 82-84% by law. I ran e85 in my GT350 for four years. That car had an ethanol sensor so I always knew my percentage and found that it never varied. I have also done the test tube test and it has always been over 80. Since my Supra runs e50 I use the tool below to calculate the proper mix and have had no problems.

Choose the gas station you like most, buy a few test kits, and test the E blend every so often to get a feel for what they usually average. Use an online ethanol calculator to calculate the mix you need for e50.

The station nearest me disclaims it can vary from e52-e85. That's a very wide range.

In states with cold winters they reduce the ethanol % to aid cold starting. In California, where the OP is from, they don't do that.
